An agonist for orphan receptors

Estrogen-related receptors share sequence similarity with estrogen receptors but bind neither estrogen nor any identified endogenous ligand — hence "orphan". They are nevertheless central to metabolism: together with the coactivator PGC-1α they drive transcription of genes for mitochondrial biogenesis and oxidative phosphorylation.

SLU-PP-322 is a synthetic agonist active across all three subtypes, and its significance is that it makes a previously unaddressable transcriptional programme pharmacologically accessible.

Research areas

  • ERR-mediated transcription and its downstream metabolic programme

  • Mitochondrial biogenesis and oxidative capacity in skeletal muscle

  • Fatty acid oxidation as a measured endpoint

  • Exercise-mimetic research — whether transcriptional programmes normally induced by activity can be induced directly

  • Nuclear receptor pharmacology, as a tool for deorphanising receptor function

An early-stage compound

SLU-PP-322 is recent and its literature is correspondingly small. There is no clinical data, and the published work is preclinical. Researchers using it should expect to be contributing to its characterisation rather than relying on an established profile.

Read alongside

AICAR reaches overlapping endpoints through AMPK rather than nuclear receptors, and the comparison between the two is a common experimental design.

Documentation and verification

Certificates of Analysis are published for tested products and batches. A certificate is a batch-specific record: it reports what was measured, by which method, on a stated date, against a batch identifier. It is evidence about the sample analysed at the time of analysis, and it is silent on attributes it does not list and on what has happened to the material since.

The batch number is the field that connects the document to a physical vial. Where the identifier on a vial does not appear on a certificate, the certificate does not describe that vial, however thorough the analysis behind it was. Batch records can be searched directly, and the guides below explain what each field establishes and what it does not.
